Q. When did we see it on the show?
A. In Episode 5 of Season One of Resurrection.
Dexter has gone out on a date with Mia (Lady
Vengeance), whom he believes may have a moral code similar to his own,
in which she only kills sexual predators.
But while at a wine bar, she reveals that her public reputation of being a moral avenger is bogus, and that she kills simply because she likes killing.
This changes everything for Dexter, and she goes from being a potential partner to being a candidate for his kill table.
During a break, Dexter goes out to his car,
which is parked in a paid lot, and retrieves his hypodermic needle and a
bottle of M99, which he is initially planning to use on Mia.
But then he realizes that after killing
Lowell and Gareth, if he kills another member of Prater's serial killer
club, the group is likely to realized that he , the newcomer, is
responsible.
So he changes strategy, and decides to get rid of her in a non-lethal way.
In the next scene, he rejoins her out on the street, where they walk & talk, Then she becomes disenchanted with him after he refuses to share one of his kills with her.
Q. What is it actually in real life?
A. An actual paid parking lot.

Where can I find it in real life?
A. This parking lot is located at 410 Lafayette St., in Manhattan.
This is the same street where Dexter & Mia are seen walking & talking, except that they are walking on the east side of the street, and this parking lot is located on the west/northwest side of the street.
It's
tucked away a narrow space between two tall buildings, less than 100
feet west of the Public Theater that Dexter & Mia pass while they
are walking.
According to Google Maps, the business name of the lot is Park-it Lafayette St. Parking, LLC.

Q. How the heck did you figure out where it was?
A. One of my most useful tools is Photoshop, which allows me to lighten, sharpen, enlarge and enhance images.
Often, this means that I can take a screencap that looks dark and
worthless i its original form, and enhance it to the point where it
brings out details you would otherwise never see.
That was the case with the screencap at the top of this page. After greatly enlarging the image, I spotted a small white sign (the 4th sign in a row below the larger main Parking sign) sign, which read 410 Lafayette.
Since I had already discovered Lafayette
Street from the scene where Dexter and Mia are walking, all I had to do
was look at a StreetView of 41O Lafayette, to verify that it was the same parking lot we see this scene.
