After
killing Nick (who had killed Brother Sam), Dexter has a crisis of
conscience and temporarily loses his bearings. He imagines his dead
brother, Brian (the Ice Truck Killer), in the same way that he usually
imagines his father Harry - as his invisible companion.
Dexter
takes off on a wild road trip to Nebraska (with Brian), to kill Jonah Mitchell, and
to contemplate ditching Harry's Code for the unstructured, murderous
abandon offered by Brian.
By the end of that trip, and after he has spared Jonah Mitchell and finally dumped Brian, we see him driving back from Nebraska.
He spots
Harry on the side of the road, hitchhiking, and stops to pick him up -
symbolizing his rejection of Brian's evil influence, and his
reconciliation with Harry's Code.
Harry says "Welcome home, Dexter", gets into his car, and they drive off together, as the episode ends.

A. A public street - but nowhere near Florida

Where can I find it in real life?
A. They shot this scene in Altadena, CA, on a stretch of Oak Grove Drive, just south of Yucca Lane.
Dexter is driving east, on the south side of Oak Grove, when he picks up Harry on the shoulder of the eastbound exit to Yucca - a street which runs above Oak Grove.
It's on the Altadena/Pasadena
border, and was listed on the Pasadena shoot sheet as "Oak Grove Dr,
from N Windsor Ave to Linda Vista".
They put up two fake signs: One, reading "Miami City Limit",
was put up on the right (south) side of the offramp, where Harry was
standing. The other, reading "South - Interstate 95", was put up on
the left (north) side, on the divider separating the offramp from Oak Grove
Drive.
While they were in the general area, they filmed the scene of Jonah Mitchell's house, down in South Pasadena.
If this location looks familiar, you might recognize it from the Disney movie "PROM", even if you've only seen the trailer for that movie.
In the "PROM" trailer, an unlucky teenager hangs a big banner from a bridge, asking a girl to go with him
to the prom. But seconds before she arrives to see it, a large
truck suddenly comes roaring down the road and rips his banner to
shreds.
They shot that scene on this exact same stretch of road, in Pasadena.

A. I recognized this location from when I originally tracked it down for Disney's Prom, just a few months ago (Plus, it was listed in the Pasadena shoot sheet as a Dexter filming location.)
